Work History

Research Intern (Mercer Lab)

Base mRNA Facility | Australian Institute of Bioengineering and Nanotechnology, University of Queensland
06.2023 - 06.2024
  • Conducted library preparation from 9 g-Block PCR products using the Rapid Barcoding Kit (RBK114.24) to compare the error rates with plasmid constructs for in vitro transcription of mRNA
  • Contributing in Developing a script for poly(A) tail analysis of plasmid and g-Block constructs, contributing to enhanced RNA stability and quality assessment
  • Utilized ONT Direct-cDNA sequencing to prepare libraries from eGFP plasmid, conducting comprehensive analyses including poly(A) tail length, error rates, and primary read lengths, crucial for optimizing mRNA vaccine formulations
  • Analyzed error rates and poly(A) tail characteristics of HA and MLI g-Block libraries prepared using the ONT ligation sequencing kit (NBD114 Kit), further refining sequencing protocols for improved vaccine efficacy
  • Optimized ONT Direct-cDNA sequencing protocols using Huh-7 Total RNA cells transfected with eGFP, establishing methods for quantifying mRNA integrity post-transfection and developing an anchored primer for poly(A) tail length analysis
  • Applied Bioinformatics Analytical pipeline to perform the analyses as well as tools such as snapgene and IGV.

Pharma research intern

Society for Health and Medical Technology | India
08.2021 - 01.2023
  • Developed a database for symptom and disease-based diagnosis support

Coursework

  • Good Clinical Practice by NIDA Clinical Trials Network
  • Bacterial Genomes I: From DNA to Protein function using Bioinformatics by Wellcome Connecting Science
  • Bacterial Genomes III: Comparative Genomics Using Artemis Comparison Tool(ACT) by Wellcome Connecting Science

Projects

  • Genome annotation using a series of bioinformatics tools Research project – “How safe and efficient are mRNA vaccines?”. I focused on the safety and immunogenicity of mRNA vaccines in comparison to conventional vaccines for three different diseases- Zika, Rabies and Covid-19.
  • Scientific News and Views Article- “Caspase-9 cleavage managed by Caspase-3 leads to full activation of the apoptosome.
  • Conducted Lab experiments and communicated my research findings in a scientific report. Two of them include- “Immobilized metal affinity chromatography (IMAC) to purify Hexahistidine tagged GFP protein from transformed E.coli XL1 cells.” And “isolation and purification of mitochondria from the meristem of a cauliflower”.
  • Research poster- Explored potential in vivo and in vitro experiments for a fictional drug with apoptotic properties similar to caspase-3, demonstrating its potential for decreased tumor growth in pancreatic cancer.
